Large Volume Tris And DPBS Buffer Preparation Using Flexsafe® Pro Mixer

Tris buffer is commonly used in downstream steps like pH equilibration, Tangential Flow Filtration and final formulation. DPBS is a buffer solution used in downstream processes to maintain a physiological pH range during protein purification, filtration or final formulation. DBPS buffer is often prepared in smaller volumes of concentrated solution, 10X or 20X ready for final dilution prior to use for space saving, or for in-line conditioning or in-line dilution used in downstream process intensification. The DPBS is thus tested at 20X in a 3,000 L system as a worse case.

Such sinking powders can be challenging to mix efficiently as they tend to accumulate on the bottom and in the corners of the bag.

This application note demonstrates the ability of the Flexsafe® Pro Mixer to mix and dissolve efficiently sinking buffer powders at large scale, making the buffer preparation step quick and easy to perform up to 3,000 L with mixing time as fast as 7 min to make 3,000 L of 1X DPBS buffer.
