A Simple, Three-Step Approach To Optimize The Fed-Batch Process For mAb Production

This article describes a straightforward way to achieve a high-performing fed-batch process.

Identifying some key items about your cell line from the beginning, such as the nutrients it needs over the culture time, impact of metabolites, and critical osmolality, can help to rapidly design a simple daily feeding regimen to perform at production scale.

We developed a fed-batch process for a CHO-K1 GS-knockout cell line (GS -/-) expressing IgG1.

The goal of the process was to overcome workflow bottlenecks that negatively affect productivity and cell growth. The strategies were developed based on a simple, three-step approach where feeds were added as a daily bolus.

Cells were grown in HyClone™ brand ActiPro™ medium and fed with HyClone™ brand Cell Boost™ 7a and 7b. The optimized feed regimens were defined in shake flasks (SF) and scaled up to bioreactor (BR) experiments.
